چکیده

Drugs and solutions All tissues were perfused and maintained with KRB solution containing (mmol/liter): 120.35 NaCl, 15.5 NaHCO3, 5.9 KCl, 1.2 MgCl2, 1.2 NaH2PO4, 2.5 CaCl2, and 11.5 glucose. The KRB solution warmed to a physiological temperature of 37 ± 0.2°C and bubbled with a mixture of 97% O2–3% CO2. For experiments using external solutions with zero [Ca]o, CaCl2 was omitted and 1 mM EGTA was added to the solution. NNC 55-0396 and TTA-A2 were purchased from Alomone Labs, nicardipine was purchased from Sigma-Aldrich, thapsigargin, isradipine, and ryanodine were purchased from Tocris Bioscience, and multiple batches of Xestospongin C (XeC) were purchased from Cayman Chemical.

Clustering of Ca2+ transients in interstitial cells of Cajal defines slow wave duration

Interstitial cells of Cajal (ICC) in the myenteric plexus region (ICC-MY) of the small intestine are pacemakers that generate rhythmic depolarizations known as slow waves. Slow waves depend on activation of Ca2+-activated Cl- channels (ANO1) in ICC, propagate actively within networks of ICC-MY, and conduct to smooth muscle cells where they generate action potentials and phasic contractions. Thu...

Effect of mibefradil on sodium and calcium currents.

Interstitial cells of Cajal (ICC) generate the electrical slow wave. The ionic conductances that contribute to the slow wave appear to vary among species. In humans, a tetrodotoxin-resistant Na+ current (Na(V)1.5) encoded by SCN5A contributes to the rising phase of the slow wave, whereas T-type Ca2+ currents have been reported from cultured mouse intestine ICC and also from canine colonic ICC. ...

Spatial and temporal mapping of pacemaker activity in interstitial cells of Cajal in mouse ileum in situ.

Spontaneous electrical pacemaker activity occurs in tunica muscularis of the gastrointestinal tract and drives phasic contractions. Interstitial cells of Cajal (ICC) are the pacemaker cells that generate and propagate electrical slow waves. We used Ca(2+) imaging to visualize spontaneous rhythmicity in ICC in the myenteric region (ICC-MY) of the murine small intestine. ICC-MY, verified by colab...

C-Kit expression in the gallbladder of guinea pig with chronic calculous cholecystitis and the effect of Artemisia capillaris Thunb on interstitial cells of Cajal

Objective(s): To study the c-Kit expression in the gallbladder of cholesterol lithogenic guinea pig model and the effect of Artemisia capillaris Thunb on interstitial cells of Cajal (ICCs). Materials and Methods:A total of 45 guinea pigs were randomly assigned into three groups: the control group (guinea pigs fed a standard diet, normal group); the model group (guinea pigs fed a cholesterol gal...

Function of interstitial cells of Cajal in the rabbit portal vein.

Interstitial cells of Cajal (ICCs) were identified in the intact fixed media of the rabbit portal vein (RPV) using c-kit staining. The following experiments were performed using single cell preparations of the enzyme-dispersed vessel. Surviving contacts between the processes of single ICCs and the bodies of smooth muscle cells (SMCs) were observed in electron micrographs and by confocal microsc...
